Maintaining sterility is a central theme in cell culture work, whether you are growing cells in a Cell Culture Dish or maintaining lines within a Cell Culture Flask. Contamination can arise from many sources, including airborne particles, tools, gloves, and even the operator themselves.
One of the basic practices is to always wear appropriate personal protective equipment (PPE) — including gloves and lab coat — and to regularly disinfect glove surfaces with ethanol. Ensuring that the work area and incubator are thoroughly cleaned before use helps create a barrier between your cultures and environmental contaminants.
Sterile handling techniques extend to how you open and close vessels. Never open a flask or dish until the instant you are ready to use it, and avoid placing the cap lid on the bench where it can collect dust or microorganisms. When handling media, use sterile pipettes and never reuse disposable tips between different cultures.
In some scenarios, air quality control within the workspace — such as using HEPA-filtered hoods and keeping incubator doors closed except when necessary — also contributes to reducing contamination risk. Many labs follow strict cleaning schedules that include wiping down all contact surfaces with disinfectants before beginning any culture work.
Another best practice is minimizing talking, coughing, or other actions that can project droplets or shed skin cells into the sterile field. Planning your steps beforehand and performing tasks quickly and efficiently reduces the time that vessels remain open, lowering chances for contaminants to enter.
